Protein Wnt-10b (formerly Wnt125) is a protein that in humans is encoded by the WNT10B gene.6789

The WNT gene family consists of structurally related genes that encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis.

This gene is a member of the WNT gene family. It may be involved in breast cancer, and its protein signaling is, it is presumed, a molecular switch that governs adipogenesis. Gain-of-function of Wnt10b in mouse hearts has shown to improve cardiac tissue repair after myocardial injury, by promoting coronary vessel formation and attenuating pathological fibrosis.10 This protein is 96% identical to the mouse Wnt10b protein at the amino acid level. This gene is clustered with another family member, WNT1, in the chromosome 12q13 region.9

A study published in August 2025 revealed another role of Wnt10b: it helps cancer cells resist stress. When a tumor cell divides too quickly, its DNA is at risk. Wnt10b acts as a stabilizer: it prevents the internal filaments (microtubules) from going at a too intense rate, avoiding improper chromosome segregation. As a result, the cancer cell survives.11

Chromosomal instability can drive dangerous tumor evolution, Wnt10b pathway is actively implicated in limiting the instability. Cancer cells use this pathway as a shield to survive the instability they create, preventing an excessive DNA damage and division errors. This makes Wnt10b a potential target for new cancer therapies. Wnt10b inhibition combined with chemotherapy could therefore become a new therapeutic strategy.12
